DNA Sequencing and Analysis Facility

 

The DNA Sequencing and Analysis Facility is designed to provide genetic analysis services to scientists at the University of Minnesota and in the community. The DNA Sequencing and Analysis Facility has state-of-the-art instrumentation and experienced staff for comprehensive solutions to your genetic analysis needs.

Please fill out online request forms for sequencing, fragment analysis, and plasmid purification. After completing form, print and submit with your samples.

Results will no longer be emailed. They will be posted on the online ordering system; go to "My Account" tab and then the "Result Files" section on the right next to "History". You will receive an email after your entire order has been posted. Please contact the lab at 612-625-7736 if you have not received an email within 72 hours of order submission. If any samples need to be reprocessed, an email will not be sent until the entire order is complete.

SUMMARY OF SERVICES PROVIDED BY THE FACILITY INCLUDE

Sanger Sequencing
The BMGC DNA Sequencing and Analysis Facility utilizes capillary electrophoretic DNA analyzers for quick turnaround on small and high-throughput projects.  The facility currently has one 96 capillary ABI 3730xl that use ABI BigDye Terminator version 3.1 chemistry.

Next-Generation Sequencing
BMGC has the GS FLX System, an ultra-high-throughput, next-generation sequencing platform capable of generating up to 400,000 long reads with exceptional accuracy in a single run. GeneMapper® 
An ABI 3730xl capillary instrument is used for fragment analysis/genotyping using GeneMapper® Software.

Plasmid Preparation
BMGC manually extracts DNA from plasmids to a sequence-ready template.

Agilent Bioanalyzer 2100
The BMGC has an Agilent Bioanalyzer 2100 available for analysis of DNA, RNA, proteins, and cells. DNA and RNA chips are available for purchase through the lab.

Real Time - PCR
An ABI 7900HT is available to researchers for customized projects. Please contact Trianna Full prior to starting your project at 612-625-7736 for details. The 7900HT has a wavelength detection of 500-660 nm, allowing for multiple chemistry detection, including TAQMAN® and SYBR® Green. Applications include real-time PCR, SNP detection and +/- assays. The 7900HT is equipped with robotic capabilities for multiple plate scanning.

Hydra pipetting stations
The BMGC has two 96-well Robbins Scientific Hydra pipetting stations. 

NanoDrop-8000

The NanoDrop ND-8000 is a full-spectrum (220-750nm) spectrophotometer that measures 2 ul samples with high accuracy and reproducibility. It utilizes a patented sample retention technology that employs surface tension alone to hold the sample in place eliminating the need for expensive cuvettes and other devices and allows for clean up in seconds. The ND-8000 also has the capability to measure highly concentrated samples without dilution (50X higher concentration compared to standard specs). There is a $2.00 charge on every visit for up to 8 samples. For more than 8 samples, there is a $0.50 charge on each additional sample.
