Whole Genome Sequencer
Sample Preparation
DNA Quality: DNA should be purified by column or gel purification protocols. Samples providing A260/A280 of > 1.8 normally provide optimal results. A number of commercially available kits will yield DNA of sufficient quality and purity for sequencing. One µL of each DNA sample should be subjected to electrophoresis on a 2% agarose gel to evaluate for concentration and quality. A hardcopy image of the EtBr-stained gel should be included with the sample(s), or sent by email. Please include a 1 kb ladder as a reference and clearly identify the contents of each lane.
DNA Quantity: For genomic sequencing projects (high molecular DNA), 10 µg of DNA provided at a concentration of > 300 ng/µL is normally adequate. For low molecular weight DNA: 3 µg, conc > 50 ng/uL, A260/A280 of > 1.8 is requested.
For other sample types please contact BMGC-Sequencing and Analysis Facility for input DNA requirements.
Sample Submissions: Samples can be provided in water or TE buffer at a minimum volume of 10 µL. Individual samples should be provided in a 1.5 mL tube labeled with content DNA and concentration. A completed Sample Description Form must be included with every sample submitted to the BMGC Sequencing and Analysis Facility.
Shipping Instructions
Packaging of Samples: Up to six 1.5 mL sample tubes should be placed into a 50 mL disposable screw-top centrifuge tube (Falcon tube) for shipping. We recommend sealing caps on individual tubes with parafilm. To prevent the sample tubes from moving during shipping, please pack the tubes with a paper towel/tissue paper.
Shipping: Samples should be frozen and shipped on dry ice. We recommend placing the 50 mL tube(s) inside of a plastic bag prior to placing on dry ice. Samples should be shipped via an express (next day) shipping carrier to: BMGC-Sequencing and Analysis Facility, Room 20 Snyder Hall, 1475 Gortner Ave., St. Paul, MN 55108
